Devon and Cornwall Police said it was called at 08:40 GMT on Sunday after a car entered the water opposite the lifeboat centre in Dartmouth.
The Maritime and Coastguard Agency said the ambulance, fire, RNLI and the HM Coastguard Rescue Helicopter were all called to the scene.
The force confirmed a man in his 80s was pronounced dead at the scene with police divers recovering his body and the vehicle from the river.
Police said a file has been passed to the coroner and the death is not believed to be suspicious.
